Cryopreservation of Malus shoot tips and subsequent plant regeneration

چکیده
The aim of the present study was to investigate the importance of the preculture conditions on the survival of apple (Malus domestica Borkh., cv. Romus4 and rootstock M106) shoot tips cryopreserved using a combined droplet vitrification procedure. Preculture was critical for the survival of apple shoot tips after cryopreservation. Among the different tested sugars and sugar alcohols (glucose, sucrose, mannitol and sorbitol) sucrose treatment for 24 h was the most efficient one. After thawing and transfer of shoot tips on recovery medium the highest regrowth rates (68% for cv. Romus4 and 62% for the rootstock M106) were achieved after using the PVS2 vitrification solution. Plants regenerated from cryopreserved shoot tips did not displayed any sign of morphological alteration or abnormalities in growth in comparison with control plants.

Preservation of In Vitro Grown Shoot Tips of Potato (Solanum tuberosum L.) by Different Methods of Cryopreservation
Cryopreservation has been recognized as a practical and efficient tool for long-term storage of vegetatively propagated plants. This study was conducted to investigate the effects of sucrose concentration, hardening temperature and different cryopreservation methods on the survival rate of potato shoot tips after cryopreservation. Excised shoot tips of in vitro plantlets of potato cultivars, At...

The present study demonstrates the potential of nutrient-alginate encapsulation of shoot tips of sunflower, Helianthus annuus (hyb. Azargol) for synthetic seed technology, which could be useful in germplasm distribution and exchange. Shoot tips from in vitro shoot cultures derived from mature seed explants were encapsulated in 3% sodium alginate and 100 mM CaCl2. 2H...
متن کاملDevelopment of cryopreservation for Loxocarya cinerea---an endemic Australian plant species important for post-mining restoration.
We report the development of a cryopreservation protocol for the endemic Western Australian plant species Loxocarya cinerea (Restionaceae). Shoot tips from two genotypes, SXH404 and SXH804, were cryopreserved using the droplet-vitrification technique. Control explants, which were cryoprotected, but not cooled, showed regeneration for both genotypes (SXH404, 22.1 +/- 5.9%; SXH804, 67.7 +/- 9.6%)...

Citrus genus is known as one of the world's major economic agricultural products. In the current research, a method is reported for artificial seed production technology by shoot tips in Citrus sinensis.var. Thamson navel. Alginate is one of the most efficient gelling agents in the production of capsule coating in artificial seeding technique. In the current research, impact of different consti...
متن کاملCryopreservation of in vitro-grown shoot tips of papaya (Carica papaya L.) by vitrification
Shoot tips from 4to 6-week-old in vitro grown plantlets of six different cultivars of papaya were cryopreserved by vitrification. Shoot tips were treated in a 2 ml cryo-tube filled with a solution of 2 M glycerol and 0.4 M sucrose at 25°C for 20 min, then dehydrated with 1 ml pre-cooled vitrification solution at 4°C for 60 min. Treated samples were kept in liquid nitrogen for at least one week....
